D6 off (not ok)
D21 off (not of)
PC user leds 1,2 red
PG user led red
Nay ideas what's going wrong?

That is not much information. Is this out of the box? Is it plugged into a PicoZed FMC Carrier? Are you booting from QSPI, SD Card, or JTAG? What are you doing?
If this is out of the box, have you already read this?

it`s plugged into a PicoZed FMC Carrier and I'am booting from QSPI. What I don't understand are the red LEDs and why I don't have a USB connection (no amber LED).

The only time the USB-UART Link LED lights is when you actually have a link, meaning the cable has to be plugged into J6 and the PC, the driver installed on the PC, and a utility (like TeraTerm) running that is connected to the link.
The PS and PL user LEDs will turn on, even when not driven, depending on how the unused I/Os and pre-configuration pull-up settings are set. As soon as you put these into your design and control them, then they will turn on and off at your choosing.
