Hello, this depends on how you plan to do the PCB.
If you do it at home - no matter with chemicals or engraving router - I suggest you cut the hole before to start the PCB job using one of the cheap large hole drill saw like in the image below. I am sure you have no difficult to find one of these tools in any Walmart or similare warehouse for few $
If you plan to make the PCB from a service, it depends on the CAD you are using. Typically if you place rounded borders external and internal of your design in the accepted layer by the service you can have it perfectly done probably with a small overpricing for the extra process.
